I just updated our technical specifications article listing our previous versions. You can find it here.
I can confirm that FME 2021.x is supported on Windows Server 2012 R2.

There should be no major changes between FME 2021 and 2022.

Mainly, I would like to know whether FME Desktop 2021.x is supported on Windows Server 2012 R2 or not?
I understand from the given link, FME 2022 is NOT supported on Windows Server 2012 R2.

That is correct, for FME Desktop.
