I would like to add a reader, which reads all files (of a specific type) in one folder. Is this possible in FME?

yes you can, when you add a reader, choose the format, next would be dataset. There you can hit a + button  (advanced browser).

 

Now you can choose folders (with option to look into subdirectories)
